MOLECULAR GENETIC ANALYSIS OF DOUBLE- STRAND DNA BREAKS IN SPERM

Double-strand DNA breaks are the most severe damage to the genome and the main genetic cause of a cell division delay despite their relatively low frequency of occurrence. After fertilization, the sperm with double-strand DNA breaks poses an increased risk of delay in the zygote development and its death before and after implantation (the “ingrowth” of an embryo into the uterine mucosa), which manifests itself in the form of infertility and pregnancy loss. Indications for the study: infertility for unknown reasons and habitual pregnancy loss in the case of normal values ​​of sperm DNA fragmentation.

Double-strand breaks in sperm DNA occur for a number of reasons, including defects in the process of formation and development of male germ cells, the effects of radio- and chemotherapy, and environmental mutagens. Even at low values of total DNA fragmentation, spermatozoa may have an increased level of DNA double-strand breaks. According to the data obtained, in approximately 20% of men with the normal indices of total DNA fragmentation, the indices of double-strand DNA breaks in spermatozoa significantly exceed the norm. The test we offer detects double-strand DNA breaks in every sperm. We perform sperm testing using a neutral version of the comet assay — the only method that allows the direct counting of DNA double-strand breaks.
